Gavreto (Pralsetinib) is a prescription medication used to treat adults with advanced stages of a form of lung cancer called non-small cell lung cancer (NSCLC). This drug is only used if the cancer is present with a specific rearrangement in a gene called rearranged during transfection (RET) and if you have not been previously treated with another RET inhibitor medicine. The active ingredient in Gavreto is Pralsetinib. Pralsetinib belongs to a class of drugs called RET inhibitors. It works by slowing or stopping the growth of cancer cells.

We currently offer the brand Gavreto from Canada manufactured by Hoffman–La Roche in the strength of 100 mg capsules. The generic Pralsetinib is not available.

Get medical help immediately if you have any of these signs or an allergic reaction while taking Gavreto. The most common side effects of Gavreto may include the following:
• Constipation;
• Cough;
• Edema;
• Muscle pain;
• Tiredness

Contact your doctor right away if you have any of the severe side effects, including:
• New or worsening cough;
• Severe headache or dizziness;
• Signs of unusual bleeding like nosebleeds, bruising, abnormal vaginal bleeding, bleeding gums;
• Wounds that will not heal

These are not all of the side effects of Gavreto. Always contact your doctor if you notice any unusual symptoms.

You should not use Gavreto if you are allergic to Pralsetinib or any ingredient in Gavreto. Before you start using this medication, tell your doctor about your medical history, especially if you:
• Are breastfeeding or planning to breastfeed;
• Are pregnant or planning to be pregnant;
• Have bleeding problems;
• Have ever had tuberculosis;
• Have high blood pressure;
• Have lung or breathing problems other than lung cancer

Tell your doctor about all your medications, including prescription, OTC, herbal supplements, and vitamins. There may be drug interactions between Gavreto and other medicines like ritonavir, ketoconazole, nefazodone, telithromycin, etc. Avoid St. John’s wort, rifampicin, carbamazepine, or phenytoin with Gavreto. Tell your doctor if you are pregnant or planning to become pregnant. Gavreto may harm an unborn baby. You should avoid becoming pregnant when using this drug. Use highly effective nom-hormonal contraception while using this drug and for at least two weeks after stopping treatment. Males with female partners of childbearing potential should use effective contraception during treatment and for 1 week after completion of treatment. Inform your doctor if you are breastfeeding or planning to breastfeed. It is not known if Gavreto passes into breast milk or if it may affect a nursing infant. You should not breastfeed during treatment with this medicine and for at least 1 week after the last dose. Use Gavreto precisely as prescribed by your doctor. Swallow the capsules whole with a glass of water on an empty stomach. Do not eat for at least two hours before and at least one hour after taking Gavreto. If you vomit after taking a dose, do not take an extra dose. Continue with your regular dosing schedule. Avoid drinking grapefruit juice or eating grapefruit or Seville oranges while using this drug. Gavreto may cause fatigue. Avoid driving or using machines until your symptoms resolve. Store Gavreto in its original package, away from moisture. Keep out of the sight and reach of children.
